PANCRELIPASE

Chemistry - Pancrelipase contains pancreatic enzymes, primarily lipase but also amylase andprotease and is obtained from the pancreas of hogs. Each mg of pancrelipase contains not less than24 USP units of lipase activity, not less than 100 USP units of protease activity, and not less than100 USP units of amylase activity. When compared on a weight basis, pancrelipase has at least 4times the trypsin and amylase content of pancreatin, and at least 12 times the lipolytic activity ofpancreatin.

Storage, Stability, Compatibility

Viokase® preparations should be stored at room temperaturein a dry place in tight containers. When present in quantities greater than trace amounts, acids willinactivate pancrelipase.

Pharmacology - PANCRELIPASE

The enzymes found in pancrelipase help to digest and absorb fats, proteins andcarbohydrates.
Uses, Indications - Pancrelipase is used to treat patients with exocrine pancreatic enzyme deficiency. It may also be used in the attempt to test for pancreatic insufficiency secondary to chronicpancreatitis.
Contraindications/Precautions - Pancrelipase products are contraindicated in animals who arehypersensitive to pork proteins.

Adverse Effects, Warnings

High doses may cause GI distress (diarrhea, cramping, nausea).
Overdosage - Overdosage may cause diarrhea or other intestinal upset. The effects should betemporary; treat by reducing dosage and supportively if diarrhea is severe.

Drug Interactions

Antacids (magnesium hydroxide, calcium carbonate) may diminish the effectiveness of pancrelipase. Cimetidine (or other H2 antagonists) may increase the amount ofpancrelipase that reaches the duodenum.
